# <b>Tissue Clearing</b>

Classical plant tissue clearing method.

# Procedure

1. Dissolve chloral hydrate with ddH<sub>2</sub>O in a glass vial by gently stirring at room temperature.

- Generally it will be fully dissolved within 30 mins;
- To speed up dissolution, heat to no more than 37&deg;C;

2. Add 1 mL glycerol and mix well after the chlorol hydrate was fully dissolved.

3. Store this clearing solution at room temperature, protected from light.

- The resulting product is a viscous, syrup-like liquid.

4. Immerse the sample in this clearing solution and leave it at room temperature until the tissue becomes transparent.

- The clearing process may takes from several hours to overnight, depend on the sample type.
